A. Narrative report of Research, Teaching, and Clinical Contributions


My research interests centers on the genetic and epigenetic factors that play a role in shaping the architecture of the brain. One area of concentration has been on the investigation of animal models of the differences that have been noted between the brains of dyslexics and those of normal readers. I am interested in the changes in the brain that accompany minor neocortical malformations that are either induced or occur spontaneously. Using congenic mice, we have determined that genes on Chrs. 4 and 8 of the mouse modulate the presence of spontaneous malformations. With collaborators, we are exploring the behavioral and anatomic consequences of induced malformations by testing rats in various auditory discrimination paradigms and subsequent examination of their brains. Male, but not female rats, with induced malformations have defects in the rapid processing of auditory information that are accompanied by changes in the cells size distribution of the medial geniculate nucleus. This sex difference appears to be related to the effects of perinatal androgens. We have also found that induction of microgyria results in profound disruption of connectivity throughout the neocortex and thalamus. We are are currently interested in the effects of the knockdown of dyslexia candidate susceptibility gene (CDSG) function on the development of the cerebral cortex. In specific, we have found that in utereo electroporation of shRNA targeted against the CCSGs Dyx1c1, Kiaa0319, and Dcdc2 disrupt neuronal migration in the cerebral cortex. We are examining the specific changes in migration and how that might disrupt brain organization.

My second area of interest centers on the investigation of genetic aspects of quantitative traits in the mouse brain. Investigation of the striatum of the mouse has shown that the volume of the striatum and the number of neurons contained with it are not correlated either anatomically or genetically. These traits were separated mapped to regions of two different chromosomes using QTL analysis. We have also mapped QTLs that modulate cerebral ventricle size as well as neocortical and striatal volume in mice, and are currently investigating other quantitative neuroanatomic traits. Finally, we have also mapped QTLs that modulate the incidence of minor malformations of the cortex. We have created a large library of high quality histologic specimens that are available over the web (The Mouse Brain Library; http://www.mbl.org). We are currently in the process of creating a transcriptome map of the striatum and cerebral cortex in both young and adult mice.
